The young Bluebirds will head into the fixture in good form, following their most recent victory against Reading in last week's cup tie.
In a hard-fought encounter, Morgan Wigley's late goal proved the difference last time out, as City defeated Reading 1-0 in Leckwith.
Now searching for a third consecutive Premier League Cup victory, City will face a strong travelling outfit in Wolves, who have won both of their cup ties during the current campaign.
Ahead of kick-off, U21s Manager Darren Purse assessed the upcoming fixture, in what will be another good challenge for the young Bluebirds.
He began: "The boys are as prepared as they can be, and the form that they're in at the moment means that we should be full of confidence going into a match against what is a very good Cat 1 academy.
"We're in a good place at this moment in time, and we'll have to see how the game goes. It was an unbelievable experience for the boys who were involved in the Peter Whittingham Memorial Match on Wednesday evening, and it just shows that the Manager wants to give the younger players the opportunity.
"It's something to be thankful for, as it could well give them that extra push to want to excel and progress into the First Team set-up on a regular basis. It shows the pathway that we've got at this football club.
"We've had some big tests on the pitch, and the challenges involved for players with the amount of travelling that they've done too. Regular matches like we've had over the last few weeks are a great way to expose the boys to the demands of senior football, and I wouldn't have it any other way."
